File Copy 2 USB (Drag & Drop)
Simple file copier to USB.

Feature:
1. AutoCopy on file drop or just click copy button.
2. Auto detect removable drives on plugin or plug-out.
3. You can select which drives as destination.

Neat idea. I could maybe see this being useful if you are trying create exact copies of the same flash drive.

It can automatically/manually (if set) copy files to all drives selected I mean the checked drives.

If a new device is plugged in, it's selected as the destination, device is remove, new device is inserted, or random device is removed, and the original device is inserted and takes over a new drive letter, will it be able to detect that it was once a selected destination?

This seems like a simple program though. It could be useful, but I usually just use the filesystem, as it shows me a good indication of how much work has already been done. That visual representation is even more improved on in Windows 8 too.

No

So I'm assuming the yes is for:
"Is it based on the mapped drive letters"

You could take it up a notch by checking the definition for a specific drive. Then depending on the selected state, save it to a file so the last settings by the user stay the same.
